Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002439.5(MSH3):c.14A>T (p.Lys5Met), citing Ambry Variant Classification Scheme 2023: The p.K5M variant (also known as c.14A>T), located in coding exon 1 of the MSH3 gene, results from an A to T substitution at nucleotide position 14. The lysine at codon 5 is replaced by methionine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
