Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014989.7(RIMS1):c.4072C>T (p.Arg1358Cys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with cysteine, which is neutral and slightly polar, at codon 1358 of the RIMS1 protein (p.Arg1358Cys). This variant is present in population databases (rs765099299, gnomAD 0.009%). This missense change has been observed in individuals with clinical features of retinal dystrophy (internal data). ClinVar contains an entry for this variant (Variation ID: 952047). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
