This variant has been observed in individuals affected with X-linked hypophosphatemia (PMID: 29460029, Invitae). ClinVar contains an entry for this variant (Variation ID: 951800). This sequence change replaces leucine with arginine at codon 673 of the PHEX protein (p.Leu673Arg). The leucine residue is highly conserved and there is a moderate physicochemical difference between leucine and arginine. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt PHEX protein function.
