Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.350C>T (p.Ser117Leu), citing Ambry Variant Classification Scheme 2023: The p.S117L variant (also known as c.350C>T), located in coding exon 5 of the POLE gene, results from a C to T substitution at nucleotide position 350. The serine at codon 117 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
