NM_020975.6(RET):c.100G>T (p.Asp34Tyr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D34Y variant (also known as c.100G>T), located in coding exon 2 of the RET gene, results from a G to T substitution at nucleotide position 100. The aspartic acid at codon 34 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
