NM_000548.5(TSC2):c.320C>A (p.Ala107Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A107D variant (also known as c.320C>A), located in coding exon 3 of the TSC2 gene, results from a C to A substitution at nucleotide position 320. The alanine at codon 107 is replaced by aspartic acid, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
