This variant lies in the SCN11A gene (transcript NM_001349253.2) at coding-DNA position 2897, where T is replaced by C; at the protein level this means replaces isoleucine at residue 966 with threonine — a missense variant. Submitter rationale: This sequence change replaces isoleucine with threonine at codon 966 of the SCN11A protein (p.Ile966Thr). The isoleucine residue is moderately conserved and there is a moderate physicochemical difference between isoleucine and threonine. This variant is present in population databases (rs761443584, ExAC 0.001%). This variant has not been reported in the literature in individuals with SCN11A-related conditions.
