NM_002180.3(IGHMBP2):c.2140G>A (p.Gly714Arg) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G714R variant (also known as c.2140G>A), located in coding exon 13 of the IGHMBP2 gene, results from a G to A substitution at nucleotide position 2140. The glycine at codon 714 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
