This variant lies in the CLN5 gene (transcript NM_006493.4) at coding-DNA position 844 through coding-DNA position 845, duplicating 2 bases; at the protein level this means shifts the reading frame starting at threonine residue 283, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change results in a premature translational stop signal in the CLN5 gene (p.Thr332Argfs*5). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 76 amino acids of the CLN5 protein. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with CLN5-related conditions. This variant disrupts the C-terminus of the CLN5 protein. Other variant(s) that disrupt this region (p.Tyr392*) have been determined to be pathogenic (PMID:9662406, 20052765, 11971870, 24058541, 24038957).
