Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_003036.4(SKI):c.149A>T (p.Glu50Val), citing Ambry Variant Classification Scheme 2023: The p.E50V variant (also known as c.149A>T), located in coding exon 1 of the SKI gene, results from an A to T substitution at nucleotide position 149. The glutamic acid at codon 50 is replaced by valine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
