Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.3131A>G (p.Tyr1044Cys), citing Ambry Variant Classification Scheme 2023: The p.Y1044C variant (also known as c.3131A>G), located in coding exon 24 of the NF1 gene, results from an A to G substitution at nucleotide position 3131. The tyrosine at codon 1044 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
