NM_058195.4(CDKN2A):c.22A>G (p.Thr8Ala) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T8A variant (also known as c.22A>G), located in coding exon 1 of the CDKN2A (p14ARF) gene, results from an A to G substitution at nucleotide position 22. The threonine at codon 8 is replaced by alanine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
