This variant lies in the POLE gene (transcript NM_006231.4) at coding-DNA position 394, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 132 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The p.E132* variant (also known as c.394G>T), located in coding exon 5 of the POLE gene, results from a G to T substitution at nucleotide position 394.
