Uncertain significance for Primary ciliary dyskinesia 32 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_031924.8(RSPH3):c.-225G>A, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the RSPH3 gene (transcript NM_031924.8) at 225 bases upstream of the translation start (5' untranslated region), where G is replaced by A. Submitter rationale: In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has not been reported in the literature in individuals with RSPH3-related conditions. This variant is present in population databases (rs144832123, ExAC 0.003%). This sequence change replaces alanine with threonine at codon 68 of the RSPH3 protein (p.Ala68Thr). The alanine residue is weakly conserved and there is a small physicochemical difference between alanine and threonine.
