This variant lies in the DMD gene (transcript NM_004006.3) at coding-DNA position 4845, where G is replaced by C; at the protein level this means replaces lysine at residue 1615 with asparagine — a missense variant. Submitter rationale: This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with DMD-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). Nucleotide substitutions within the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098).
