Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.3317C>A (p.Ser1106Tyr), citing Ambry Variant Classification Scheme 2023: The p.S1106Y variant (also known as c.3317C>A), located in coding exon 20 of the DICER1 gene, results from a C to A substitution at nucleotide position 3317. The serine at codon 1106 is replaced by tyrosine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
