Uncertain significance for Pfeiffer syndrome; Hypogonadotropic hypogonadism 2 with or without anosmia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_023110.3(FGFR1):c.295T>C (p.Tyr99His), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tyrosine with histidine at codon 99 of the FGFR1 protein (p.Tyr99His). The tyrosine residue is moderately conserved and there is a moderate physicochemical difference between tyrosine and histidine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with FGFR1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
