For these reasons, this variant has been classified as Pathogenic. The current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in ASXL3 cause disease. This variant has been observed to be de novo in an individual affected with clinical features of Bainbridge-Ropers syndrome (Invitae). This sequence change creates a premature translational stop signal (p.Leu499Phefs*9) in the ASXL3 gene. It is expected to result in an absent or disrupted protein product.
