NM_000051.4(ATM):c.6146dup (p.Tyr2049Ter) was classified as Pathogenic for Ataxia-telangiectasia syndrome by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in ATM are known to be pathogenic (PMID: 23807571, 25614872). This variant has not been reported in the literature in individuals with ATM-related conditions. However, a different variant (c.6146_6147del) giving rise to the same protein effect observed here (p.Tyr2049*) has been observed in an individual affected with breast and/or ovarian cancer (PMID: 29470806). This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Tyr2049*) in the ATM gene. It is expected to result in an absent or disrupted protein product.
