Uncertain significance for Emery-Dreifuss muscular dystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001130965.3(SUN1):c.1372C>G (p.Gln458Glu), citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (rs547404802, gnomAD 0.02%). This sequence change replaces glutamine, which is neutral and polar, with glutamic acid, which is acidic and polar, at codon 458 of the SUN1 protein (p.Gln458Glu). This variant has not been reported in the literature in individuals affected with SUN1-related conditions. ClinVar contains an entry for this variant (Variation ID: 947406). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The glutamic acid amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
