Uncertain significance for Hyper-IgM syndrome type 5 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_080911.3(UNG):c.32T>C (p.Phe11Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces phenylalanine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 11 of the UNG protein (p.Phe11Ser). This variant is present in population databases (rs747684225, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with UNG-related conditions. ClinVar contains an entry for this variant (Variation ID: 947219). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
