This variant lies in the ACTN2 gene (transcript NM_001103.4) at coding-DNA position 2168, where G is replaced by C; at the protein level this means replaces glycine at residue 723 with alanine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
