This variant lies in the WNK1 gene (transcript NM_018979.4) at coding-DNA position 2162, where A is replaced by G; at the protein level this means replaces glutamine at residue 721 with arginine — a missense variant. Submitter rationale: This sequence change replaces glutamine with arginine at codon 1219 of the WNK1 protein (p.Gln1219Arg). The glutamine residue is weakly conserved and there is a small physicochemical difference between glutamine and arginine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with WNK1-related conditions.
