Uncertain significance for Autosomal recessive DOPA responsive dystonia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000360.4(TH):c.730G>T (p.Ala244Ser), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the TH gene (transcript NM_000360.4) at coding-DNA position 730, where G is replaced by T; at the protein level this means replaces alanine at residue 244 with serine — a missense variant. Submitter rationale: This sequence change replaces alanine with serine at codon 275 of the TH protein (p.Ala275Ser). The alanine residue is moderately conserved and there is a moderate physicochemical difference between alanine and serine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with TH-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
