This variant lies in the POLD1 gene (transcript NM_002691.4) at coding-DNA position 1735, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 579 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: Missense variants that disrupt the 3'-5' exonuclease (proof-reading) activity of the POLD1 protein, while not abolishing its polymerase enzyme activity, are associated with an increased risk for colonic adenomatous polyps and colon cancer (PMID: 23263490, 23447401).
