NM_006231.4(POLE):c.6504C>G (p.Asp2168Glu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D2168E variant (also known as c.6504C>G), located in coding exon 46 of the POLE gene, results from a C to G substitution at nucleotide position 6504. The aspartic acid at codon 2168 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
