This variant lies in the B4GAT1 gene (transcript NM_006876.3) at coding-DNA position 1018, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 340 with lysine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This variant has not been reported in the literature in individuals affected with B4GAT1-related conditions. This sequence change replaces glutamic acid with lysine at codon 340 of the B4GAT1 protein (p.Glu340Lys).
