Uncertain significance for Diamond-Blackfan anemia — the classification assigned by Ambry Genetics to NM_000987.5(RPL26):c.377G>A (p.Arg126His), citing Ambry Variant Classification Scheme 2023: The p.R126H variant (also known as c.377G>A), located in coding exon 3 of the RPL26 gene, results from a G to A substitution at nucleotide position 377. The arginine at codon 126 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
