Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_015450.3(POT1):c.1156C>G (p.His386Asp), citing Ambry Variant Classification Scheme 2023: The p.H386D variant (also known as c.1156C>G), located in coding exon 9 of the POT1 gene, results from a C to G substitution at nucleotide position 1156. The histidine at codon 386 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
