This variant lies in the ALDOB gene (transcript NM_000035.4) at coding-DNA position 2, where T is replaced by C; at the protein level this means replaces methionine at residue 1 with threonine — a missense variant. Submitter rationale: This sequence change affects the initiator methionine of the ALDOB mRNA. The next in-frame methionine is located at codon 40. This variant is present in population databases (no rsID available, gnomAD 0.002%). Disruption of the initiator codon has been observed in individual(s) with hereditary fructose intolerance (PMID: 8438046). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 945084).
