NM_000548.5(TSC2):c.4270G>C (p.Asp1424His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1424H variant (also known as c.4270G>C), located in coding exon 33 of the TSC2 gene, results from a G to C substitution at nucleotide position 4270. The aspartic acid at codon 1424 is replaced by histidine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
