This variant lies in the DMD gene (transcript NM_004006.3) at 18 bases into the intron immediately before coding-DNA position 1332, where C is replaced by T. Submitter rationale: Variant summary: DMD c.1332-18C>T alters a nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. 4/4 computational tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 0.0021 in 173111 control chromosomes in the gnomAD database, including 3 homozygotes. The observed variant frequency is approximately 192.7- fold the estimated maximal expected allele frequency for a pathogenic variant in DMD causing Dystrophinopathies phenotype (1.1e-05), strongly suggesting that the variant is benign. To our knowledge, no occurrence of c.1332-18C>T in individuals affected with Dystrophinopathies and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as benign.
