Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001035.3(RYR2):c.14121G>A (p.Met4707Ile), citing Ambry Variant Classification Scheme 2023: The p.M4707I variant (also known as c.14121G>A), located in coding exon 98 of the RYR2 gene, results from a G to A substitution at nucleotide position 14121. The methionine at codon 4707 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species, and isoleucine is the reference amino acid in other vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
