NM_017636.4(TRPM4):c.2840C>A (p.Ala947Asp) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A947D variant (also known as c.2840C>A), located in coding exon 19 of the TRPM4 gene, results from a C to A substitution at nucleotide position 2840. The alanine at codon 947 is replaced by aspartic acid, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
