Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000222.3(KIT):c.214G>T (p.Asp72Tyr), citing Ambry Variant Classification Scheme 2023: The p.D72Y variant (also known as c.214G>T), located in coding exon 2 of the KIT gene, results from a G to T substitution at nucleotide position 214. The aspartic acid at codon 72 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
