Uncertain significance for Bethlem myopathy 1A — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004369.4(COL6A3):c.4363A>G (p.Ile1455Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine with valine at codon 1455 of the COL6A3 protein (p.Ile1455Val). The isoleucine residue is moderately conserved and there is a small physicochemical difference between isoleucine and valine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with COL6A3-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
