Uncertain significance for Neutropenia, severe congenital, 1, autosomal dominant; Cyclical neutropenia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001972.4(ELANE):c.505C>A (p.Leu169Met), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine with methionine at codon 169 of the ELANE protein (p.Leu169Met). The leucine residue is highly conserved and there is a small physicochemical difference between leucine and methionine. This variant is present in population databases (rs749675557, ExAC 0.003%). This variant has not been reported in the literature in individuals with ELANE-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
