This variant lies in the MLH1 gene (transcript NM_000249.4) at coding-DNA position 344, where T is replaced by C; at the protein level this means replaces isoleucine at residue 115 with threonine — a missense variant. Submitter rationale: This sequence change replaces isoleucine with threonine at codon 115 of the MLH1 protein (p.Ile115Thr). The isoleucine residue is highly conserved and there is a moderate physicochemical difference between isoleucine and threonine. This variant is present in population databases (rs764120517, ExAC 0.006%). This variant has not been reported in the literature in individuals with MLH1-related conditions.
