Uncertain significance for Hermansky-Pudlak syndrome 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003664.5(AP3B1):c.2603C>T (p.Thr868Met), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine with methionine at codon 868 of the AP3B1 protein (p.Thr868Met). The threonine residue is weakly conserved and there is a moderate physicochemical difference between threonine and methionine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with AP3B1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0". The methionine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
