Uncertain significance for Left ventricular noncompaction 8 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_022114.4(PRDM16):c.1753G>A (p.Asp585Asn), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with PRDM16-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces aspartic acid with asparagine at codon 585 of the PRDM16 protein (p.Asp585Asn). The aspartic acid residue is moderately conserved and there is a small physicochemical difference between aspartic acid and asparagine. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The asparagine amino acid residue is found in multiple mammalian species, suggesting that this missense change does not adversely affect protein function.
