NM_177438.3(DICER1):c.4814C>G (p.Pro1605Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P1605R variant (also known as c.4814C>G), located in coding exon 22 of the DICER1 gene, results from a C to G substitution at nucleotide position 4814. The proline at codon 1605 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
