Uncertain significance for Jeune thoracic dystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020800.3(IFT80):c.1052C>T (p.Thr351Met), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine, which is neutral and polar, with methionine, which is neutral and non-polar, at codon 351 of the IFT80 protein (p.Thr351Met). This variant is present in population databases (rs199778285, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with IFT80-related conditions. ClinVar contains an entry for this variant (Variation ID: 942970). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site.
