NM_001943.5(DSG2):c.385G>T (p.Gly129Cys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G129C variant (also known as c.385G>T), located in coding exon 5 of the DSG2 gene, results from a G to T substitution at nucleotide position 385. The glycine at codon 129 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is well conserved in available vertebrate species; however, cysteine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
