This variant lies in the SDHA gene (transcript NM_004168.4) at coding-DNA position 646, where T is replaced by C; at the protein level this means replaces phenylalanine at residue 216 with leucine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C15"). This variant has not been reported in the literature in individuals with SDHA-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces phenylalanine with leucine at codon 216 of the SDHA protein (p.Phe216Leu).
