Uncertain significance for Ehlers-Danlos syndrome, dermatosparaxis type — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014244.5(ADAMTS2):c.2794C>T (p.Arg932Trp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine with tryptophan at codon 932 of the ADAMTS2 protein (p.Arg932Trp). The arginine residue is highly conserved and there is a moderate physicochemical difference between arginine and tryptophan. This variant is present in population databases (rs776975672, ExAC 0.01%). This variant has not been reported in the literature in individuals affected with ADAMTS2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C35").
