Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001134363.3(RBM20):c.2021A>G (p.Asp674Gly), citing Ambry Variant Classification Scheme 2023: The p.D674G variant (also known as c.2021A>G), located in coding exon 9 of the RBM20 gene, results from an A to G substitution at nucleotide position 2021. The aspartic acid at codon 674 is replaced by glycine, an amino acid with similar properties. This variant was reported in individual(s) with features consistent with dilated cardiomyopathy (Hey TM et al. Circ Heart Fail, 2019 Mar;12:e005700; Marschall C et al. Cardiovasc Diagn Ther, 2019 Oct;9:S292-S298). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
