Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_003242.6(TGFBR2):c.670C>T (p.Arg224Cys), citing Ambry Variant Classification Scheme 2023: The p.R224C variant (also known as c.670C>T), located in coding exon 4 of the TGFBR2 gene, results from a C to T substitution at nucleotide position 670. The arginine at codon 224 is replaced by cysteine, an amino acid with highly dissimilar properties. This variant was reported in an individual in a heritable thoracic disease cohort (Jondeau G et al. Circ Cardiovasc Genet, 2016 Dec;9:548-558). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
