Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000138.5(FBN1):c.843A>T (p.Glu281Asp), citing Ambry Variant Classification Scheme 2023: The p.E281D variant (also known as c.843A>T), located in coding exon 7 of the FBN1 gene, results from an A to T substitution at nucleotide position 843. The glutamic acid at codon 281 is replaced by aspartic acid, an amino acid with highly similar properties, and is located in the cbEGF-like #01 domain. This amino acid position is well conserved in available vertebrate species; however, aspartic acid is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
