This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 9595 through coding-DNA position 9596, duplicating 2 bases; at the protein level this means shifts the reading frame starting at serine residue 3200, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This variant is not present in population databases (ExAC no frequency). This sequence change results in a premature translational stop signal in the BRCA2 gene (p.Ser3200Leufs*18). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 219 amino acids of the BRCA2 protein. This variant has not been reported in the literature in individuals with BRCA2-related conditions. This variant disrupts the C-terminus of the BRCA2 protein. Other variant(s) that disrupt this region (p.Tyr3308*) have been determined to be pathogenic (PMID: 17026620, 18593900, 18607349, 22711857).
