Uncertain significance for Charcot-Marie-Tooth disease axonal type 2O — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001376.5(DYNC1H1):c.3287C>T (p.Ala1096Val), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This sequence change replaces alanine with valine at codon 1096 of the DYNC1H1 protein (p.Ala1096Val). The alanine residue is highly conserved and there is a small physicochemical difference between alanine and valine. This variant has not been reported in the literature in individuals with DYNC1H1-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
